there is no single best one. A combination of something like spybot search and destroy, ad-aware and spyware blaster should serve you well.

back it up with a good solid anti-virus.

I agree. You should combine few anti-spyware programs. I use Spybot + Spyware Doctor + Ad Aware Pro.

I wonder what makes an antispyware the best, inasmuch as all that I have seen remove the spyware and adware cookies or registry entries. Everybody seems to list whatever program they happen to be using.
There is one program that is using a different process and that is Winpooch at Sourceforge. It is a long way from being polished, but it constantly sniffs the registry, hence 'Winpooch'.

Speaking of registry changes, Spybot has a resident scanner that resides in the taskbar, and will inform you of any attempted changes to the registry. It allows you to block or allow them.

Aside from that, I use the usual Spybot + AdAware combo, and I've found Norton 360 to be a very good antivirus/anti-spyware/firewall combo for Windows Vista. It's very easy on the resources compared to older Norton products, so easy on the resources that I don't notice it at all, and it gives better protection that what I was using. (basic windows firewall + nod32.)

Norton 360 isn't free (or cheap, for that matter) but it does do really well.
My current setup stats (like anyone cares...):

    ASUS A8N32-SLI Motherboard
    AMD 4400+ Dual-Core CPU
    Windows Vista (Ultimate 32bit)
2 GB (2x1GB) Corsair XMS RAM
2x250 GB (in RAID 0) HDDs
EVGA GeForce 7950 GTX 512 MB
    Creative X-FI Fatal1ty XtremeGamer

    Also sporting a black MacBook
    Revision/Release 1
    Upgraded to 2GB RAM.

spysweeper is a hungry package though i think it suffers from memory leaks,

but the key is not to have a tool that is good at removing , but a tool which prevents them getting there in the first place, and no one tool can do that a mixiture of about 5 to cover all bases

The only ones that I don't seem to have the ability to stop, limit or control; are those that get to me via one of the Instant Messengers. Can you do anything with them?

Mcafee all in 1 pakage and adblocking seems to do job for me, but I hav tried spy sweeper, spyware doctor, AVG, Norton 360, Spybot AdAware pro, spyware blaster, etc. but all have small holes through them, and some times the don't work well.
AS my 3DS max teacher used to say, "If you can block a virus its not a virus". It means we need a program to get the unknown viruses etc.. and the known ones. May be some1 can Have time and will be smart enought to make one.

While this may not be completely spyware related, i always run CCleaner after I run spybot or something. It cleans up other temporary crap in the computer

We just can also be very careful when installing free software. A lot of free software comes with adware. That why it's free, and they warn you about that. But if you skip reading then terms and conditions, you may get a surprise after installing that software.

    :)
